Experimental Studies on the Influences of Testis upon the Allergic Changes in the Blood Vessels


Abstract
Since it has been postulated that endocrine glands may give some influences to allergic reaction, various investigations on steroid hormones have become as a target subject to clarify the mechanism of allergic phenomenon, endocrinologically.
Sex hormones are closely related to the production of antibody. Professor Yun described the diminution of anaphylactic shock in the castrated animals, prior to the inhibition of allergic changes in the muscle of the orchiectomized adult rabbits is reported. Meanwhile, the administration of testis hormone reduces the allergic changes in the stomach, orchiectomy aggravated the allergic changes in the duodenum, is reported.
The experiment has been planned to study the influences of testis upon the allergic changes in the aorta of rabbits.
Experimental Material and Method
Normal adult rabbits weighing around 2 kilograms were used for this experiment.
The rabbits were divided into the sensitized..,,, - nonsensitized and subdivided furtherly into 6 groups ¢¥¢¥j each, which consist of normal, the orchiectomized, the :. , transplanted which transplantation testis was performed after castration, the testosterone administrated which testosterone was given after castration, the transplanted - which transplantation of testis was performed without castration and the testosterone administrated which the testosterone was given without castration.
Each of the sensitized rabbits was injected 1cc of normal horse serum per kilogram of body weight into the subcutaneous tissue daily for days for 4 sensitizing and 0.1 cc of normal horse serum per kilogram of body weight was given locally into the wall of the aorta 2 weeks later.
Each of the unsensitized rabbits was given 0.1 cc of normal horse serum per kilogram of body weight into the wall of the aorta without sensitization.
The nonsensitized and sensitized rabbits were sacrificed at the end of 3, 6, 12, 24, 48, 72 hours and 1, 2 weeks after the final injection. The sections were prepared by hematoxylin-eosin, Van-Gieson stains and were submitted for microscopic examination.
Results
1. Nonsensitizing group
Grossly, the resected aorta showed swelling, mild congestion and petechiae. These findings were observed at 6-24 hours and gradually subsided at 48-72 hours in the normal, orchiectomized transplanted which tran-
splantation of testis was performed after castration, testosterone administrated which administration of testosterone was given after castration, transplanted which transplantation of testis was performed without castration and the testosterone administrated which testosterone was given with castration.
Microscopically, cloudy swelling, mild congestion and some nonspecific acute inflammatory cells were observed at 6-24 or 48 hours and subsided 48-72 hours in the whole groups of the nonsensitized rabbits.
2. Sensitized groups
Grossly, resected aorta showed cloudy swelling, congestion and hemorrhage. These changes were obser-
ved at 6-48 hours and gradually subsided in 72 hours 1 week. Such finding were aggravated at 24-48 hours.
The above changes in the orchiectomized rabbits were milder than in those of the normal, transplanted which transplantation of testis was performed after castration, testosterone administrated which testosterone was given after castration, transplanted which the transplantation of testis was done without castration and the testosterone administrated which testosterone was given without castration.
Microscopically, cloudy swelling, degeneration, congestion, hemorrhage, and inflammatory cells, neutrophiles, lymphocytes, plasma cells, and eosinophiles were observed at 6-48 hours and gradually subsided at 72 hours or 1 -2 weeks in the normal group. Such findings were
intense at 24-48 hours showing cloudy swelling, fibrinoid degeneration of blood vessel wall, congestion, hemorrhage and infiltration, of inflammatory cells.
In the orchiectomized rabbits, the above changes were much less than in those of the normal group.
In the transplanted which transplantation of testis was performed after castration and the testosterone administrated which testosterone was given after castration, the changes were similar in its intensity as in those of the normal group.
In the transplanted which transplantation of testis was done without castration and the testosterone administrated which testosterone was given without castration, the changes were found as in the normal group, but the intensity of the changes appeared somewhat aggravated that in those of the normal group.
Conclusion
Studies on the influences of orchiectomy upon the allergic changes in the blood vessel are reported.
The allergic changes in the orchiectomized group were much less than in those of the normal group.
The allergic changes in the transplanted which trans-plantation of testis was performed after castration, the testosterone administrated which testosterone was given after castration, the transplanted which transplantation of testis was performed without castration and the testosterone administrated which testosterone was given without castration were not more remarkable than in those of the normal group.
